Fast, Simple Method for Analysis of Benzodiazepines in Meconium


A novel method for the quantitation of 10 commonly prescribed benzodiazepines and/or their metabolites, 7-aminoclonazepam, clonazepam, α-hydroxyalprazolam, alprazolam, nordiazepam, diazepam, midazolam, oxazepam, lorazepam, and temazepam, in meconium was developed using IMCSzyme™ for enzymatic hydrolysis, WAX-S (DPX) tips, and LC-MS-MS analysis.

The method was linear over a range of 5 ng/g to 1000 ng/g with all correlation coefficients above 0.99. A blind study with a corresponding laboratory of 35 positive patient samples resulted in approximately 92% correlation, with 100% accuracy in qualitative results. The method established herein is characterized by simple vortexing for sample homogenation, fast and reliable IMCSzymeTM for hydrolysis in situ minimizing hydrolysis time to one hour, and a single WAX-S DPX tip for sample preparation that produces a small amount of clean, analyte rich acetonitrile to minimize sample preparation and solvent evaporation times. This method is quick yet effective.
